Bryophytes are small plants with gametophyte dominance and do not have vascular tissue and rhizome parts, the plant structure is relatively simple, mainly growing in a dark and humid environment, some moss plants grow in a desert environment. Although bryophytes do not have rhizomes in the strict sense, they have anthropomorphic stems and roots and can release spores continuously.

  • Tissue culture materials

At present, the materials we have successfully used for tissue culture of bryophytes are mainly gametophytes, sporophytes, and protoplasts of bryophytes, in addition to their reproductive organs, budding spores, free protoplasts, etc. for tissue culture to obtain the corresponding healing tissues or regenerated plants.

  • Selection of culture medium

The basic media we use for bryophyte tissue culture are MS medium, Knop medium, Benecke medium, K2 medium, Prat medium, etc. The types of basic media suitable for exosome culture and vegetative regeneration of different bryophytes vary, as do the components in the media.

  • Phytohormones

We add different ratios of phytohormones in different media to promote tissue culture according to the source and status of different explants. For example, IAA, IBA, 2,4-D, etc. may be used to promote the elongation of bryophyte stems, 6-BA is used to promote the expansion of bryophyte stem tip cells into a ball shape, and KT is used to induce the production of healing tissue.
