  • Selection of explants

  • Selection of culture medium

The choice of medium greatly influences the induction of differentiation in Bignoniaceae plants. MS, DKW, WPM, 1/2MS, B5, and N6 are all commonly used media for tissue culture of Bignoniaceae plants.

Under aseptic conditions, the induced axillary bud tips were cut and immediately inoculated in an N6 medium with appropriate BA and NAA to promote the proliferation culture of the buds.

  • Rooting culture and transplanting

When the adventitious buds inoculated on the induction medium grow small leaves, the upper stem tip is cut and transferred to the rooting medium for rooting culture. The seedlings can be transplanted to the greenhouse when they have produced a proper number of strong roots and are about 3-4 cm tall.
